KARYA TULIS ILMIAH UJI AKTIVITAS TONIKUM EKSTRAK ETANOL KULIT BIJI MELINJO (Gnetum gnemon L.) PADA MENCIT JANTAN (Mus musculus) DENGAN METODE KETAHANAN LAMA WAKTU BERENANG

ABSTRAK
Tanaman melinjo (Gnetum gnemon L.) mempunyai berbagai senyawa metabolit sekunder, senyawa tersebut dapat berfungsi sebagai bahan obat tradisional, kulit biji melinjo (Gnetum gnemon L.) memiliki kandungan senyawa aktif seperti alkaloid, flavonoid, dan tanin. Tujuan penelitian ini untuk mengetahui adanya efek tonikum pada ekstrak etanol kulit biji melinjo (Gnetum gnemon L.) dengan dosis 200mg/kgBB, 400mg/kgBB, dan 800mg/kgBB, dan untuk mengetahui dosis berapakah yang memberikan efek tonikum dibandingkan dengan kafein (kontrol positif) dosis 0,26mg sekali minum pada mencit jantan (Mus musculus). Metode penelitian ini menggunakan ekstrak etanol kulit biji melinjo (Gnetum gnemon L.) dengan dosis 200mg/kgBB, 400mg/kgBB, dan 800mg/kgBB dengan menggunakan metode ketahanan lama waktu berenang. Hasil penelitian menunjukkan bahwa ekstrak etanol kulit biji melinjo (Gnetum gnemon L.) memiliki efek tonikum pada dosis 200mg/kgBB, 400mg/kgBB, dan 800mg/kgBB, diperoleh selisih rata-rata lama waktu berenang sebesar 49,4 detik, 596,2 detik, dan 112,6 detik dibandingkan dengan kontrol positif kafein dosis 0,26mg sekali minum sebesar 513,2 detik. Kesimpulan ada aktivitas tonikum pada ekstrak etanol kulit biji melinjo (Gentum gnemon L.) pada dosis 400mg/kgBB dibandingkan dengan dosis 200mg/kgBB dan 800mg/kgBB, serta memiliki selisih rata-rata lama waktu berenang setara dengan kontrol positif kafein dosis 0,26mg sekali minum. Data ini didukung dengan One Way ANOVA didapatkan nilai sig 0,000 < 0,05 yang berarti terdapat perbedaan aktivitas tonikum yang signifikan dari dosis 200mg/kgBB, 400mg/kgBB, dan 800mg/kgBB dari ekstrak etanol kulit biji melinjo (Gnetum gnemon L.), kontrol positif (kafein) dosis 0,26mg sekali minum, dan control negative (Na CMC 0,5%).
ABSTRACT
The melinjo plant (Gnetum gnemon L.) has various secondary metabolites, these compounds can function as ingredients for trsditional medicine, the seed coat of melinjo (Gnetum gnemon L.) contains active compounds such as alkaloids, flavonoids, and tannins. Research purpose to determine the tonic effect on the ethanolic extract of the melinjo seed coat (Gnetum gnemon L.) at a dose of 200mg/kgBW, 400mg/kgBW, and 800mg/kgBW, and to determine the dose of melinjo seed coat (Gnetum gnemon L.) ethanol extract which gave the effect tonic compared with caffeine (positive control) dose at 0.26mg once taken in male mice (Mus musculus). Research method used ethanol extract of melinjo seed coat (Gnetum gnemon L.) at a dose of 200mg/kgBW, 400mg/kgBW, and 800mg/kgBW using the long endurance of swimming method. Result showed that the ethanolic extrac of melinjo seed coat (Gnetum gnemon L.) had a tonic effect at doses of 200mg/kgBW, 400mg/kgBW, and 800mg/kgBW, the difference between the average length of swimming time awas 49,4 seconds, 596.2 seconds, and 112,6 seconds compared to a positive control of 0.26mg caffeine dose once drink of 513. 2 seconds. Conclusion is that there is tonic activity in the ethanolic extract of the melinjo seed coat (Gnetum gnemon L.) at a dose of 400mg/kgBW compared to a dose 200mg/kgBB and 800mg/kgBB, and has a difference in the average length of swimming time equivalent to a positive control of caffeine at dose of 0,26mg once taken. This data is supported by One Way ANOVA, the sig value is 0.000 < 0.05, which means that there is a significant difference in tonic activity from doses of 200mg/kgBW, 400mg/kgBW, and 800mg/kgBW from the ethanolic extract of melinjo seed coat (Gnetum gnemon L.), positive control (caffeine) 0.26 dose once drink, and negative control (Na CMC 0.5%).
