KARYA TULIS ILMIAH UJI EFEK TONIKUM EKSTRAK ETANOL DAUN KEMANGI (Ocimum sanctum) PADA MENCIT TANTAN (Mus musculus)

ABSTRAK
Tonikum adalah suatu bahan yang dapat digunakan untuk menambah tenaga atau energi pada tubuh. Kandungan kimia daun kemangi flavonoid, tanin, dan saponin berkhasiat sebagai tonikum. Tujuan penelitian, mengetahui ekstrak etanol daun kemangi (Ocimum sactum) terhadap efek tonikum dan dosis paling efektif pada mencit (Mus musculus) dibandingkan dengan kontrol positif (kafein) dan kontrol negatif (CMC-Na 0,5%). Metode penelitian, True eksperimental dengan pendekatan pretes postes with control group, dengan melakukan perbedaan dosis ekstrak daun kemangi dengan dosis 100mg/kgBB, 200mg/kgBB, dan 400mg/kgBB mencit jantan, sedangkan kontrol positif (kafein) dengan dosis 100mg/kgBB dan kontrol negatif (CMC-Na 0,5%). Analisis data dengan cara observasi lama waktu lelah dari mulai mencit dimasukkan kedalam aquarium hingga mencit lelah dengan menunjukkan kepala berada di permukaan air 4-5 detik, setelah itu diberi ekstrak etanol lalu mencit direnangkan kembali. Hasil penelitian, (1) Ekstrak etanol daun kemangi (Ocimum sanctum) memiliki pengaruh efek tonikum baik pada dosis 400mg/kg BB dengan rata-rata lama berenang selama 1.004 detik (16 menit 44 detik), dosis 200mg/kgBB dengan rata-rata lama berenang selama 810 detik (13 menit 30 detik), dosis 100mg/kgBB dengan rata-rata lama berenang selama 706 detik (11 menit 46 detik) pada mencit jantan. (2) Dosis ekstrak etanol daun kemangi yang paling efektif tersebut adalah 400mg/kgBB mencit jantan. Perlakuan kontrol positif (kafein) diperoleh hasil dengan rata-rata lama berenang selama 1.049 detik (17 menit 29 detik) dan kontrol negatif (CMC-Na 0,5%) dengan rata-rata lama berenang selama 335 detik (05 menit 35 detik). (3) Data tersebut didukung oleh hasil statistik One Way Anova yang menunjukkan nilai signifikan 0,000 yang menunjukkan adanya perbedaan nilai signifikan antara perlakuan dosis ekstrak dengan kontrol positif dan kontrol negatif.
ABSTRACT
Tonic is a substance that can be used to increase energy or energy in the body. The chemical content of basil leaves is flavonoids, tannins, and saponins which are efficacious as tonics. The purpose of the study, was to determine the ethanolic extract of basil (Ocimum sanctum) leaves on the tonic effect and the most effective dose in mice (Mus musculus) compared to positive control (caffeine) and negative control (CMC-Na 0.5%). The research method, True experimental with a pretest posttest approach with control group, by doing different doses of basil leaf extract at a dose of 100mg/kgBW, 200mg/kgBW, and 400mg/kgBW male mice, while the positive control (caffeine) at a dose of 100mg/kgBW and control negative (CMC-Na 0.5%). Analysis of the data by observing the length of time tired from the start of the mice being put into the aquarium until the mice were tired by showing the head was on the surface of the water 4-5 seconds, after that given ethanol extract and then the mice were swimming again. The results of the study, (1) The ethanolic extract of basil leaves (Ocimum sanctum) had a good tonic effect at a dose of 400mg/kg BW with an average swimming time of 1,004 seconds (16 minutes 44 seconds), a dose of 200mg/kgBW with an average length of time. swimming for 810 seconds (13 minutes 30 seconds), a dose of 100mg/kgBW with an average length of swimming for 706 seconds (11 minutes 46 seconds) in male mice. (2) The most effective dose of ethanol extract of basil leaves was 400mg/kgBW male mice. The positive control treatment (caffeine) resulted in an average swimming length of 1,049 seconds (17 minutes 29 seconds) and a negative control (CMC-Na 0.5%) with an average swimming length of 335 seconds (05 minutes 35 seconds). (3) The data is supported by the statistical results of One Way Anova which shows a significant value of 0.000 which indicates a significant difference in the value of the extract dose treatment with positive control and negative control.
